6-Thioguanosine (6-thio-G) has applications in ribozyme and siRNA research, as well as in RNA-protein interactions. The removal of the silyl protecting group without interfering with the sulfur is critical. This is removed1 cleanly by triethylamine trihydrofluoride in DMSO but t-butylammonium fluoride (TBAF) leads to degradation of the thio-nucleotide analogue and should not be used.

Usage
Coupling: 12 minute coupling time recommended. Use 0.02M Iodine to avoid degradation and Ac-C (10-3015).
Deprotection: Treat column with 1M DBU in ACN for 4 hrs at RT, rinse and dry support. Then deprotect with 50mM NaSH in AMA at 65°C for 10 minutes. Complete using RNA deprotection. See Technical Bulletin for details (http://www.glenresearch.com/Technical/TB_RNA_TBDMS_Deprotection.pdf).
