Description

Locked Nucleic Acid (LNA) is a modified ribonucleic acid containing a methylene bridge connecting the 2’-oxygen and 4’-carbon atoms. Oligonucleotides modified with (LNA), are prominent for hybridization assays and probes due to enhanced thermal stability towards complementary oligos, without compromising base pairing specificity. Each LNA modification can increase the melting temperature of a duplex by 2-8 °C. LNAs are commonly employed in antisense oligonucleotides ASOs as gapmers or mixers.

Our locked analog or LA CPGs are designed to produce 3’-LNA modified oligonucleotides. The pyrimidine bases are thymidine and 5-methylcytosine, rather than uracil and cytosine, respectively.

Usage

  • Coupling: This support should be used in a manner identical to normal protected nucleoside support since it contains the DMT group.
  • Deprotection: Deprotect using the protocol required by the nucleobases.
