Diluent: Anhydrous Acetonitrile
Coupling: A 3 minute coupling time is recommended.
Deprotection: Use AMA and deprotect as required by the nucleobases. With ammonium hydroxide, deprotect at 65 °C/2h or RT/17h for excellent results; 55 °C/17h will yield acceptable results. A minor side reaction leading to a non-fluorescent impurity during deprotection with ammonium hydroxide can occur when using a fast cleaving support, e.g., Glen UnySupport FC. Treatment of the support prior to cleavage/deprotection with a hindered base, such as 10% diethylamine in ACN, for 2-3 minutes will prevent this side reaction.
