Diluent: 60% pyridine/40% Acetonitrile (fully dissolve in pyridine first, see notes below*) *For a 0.05M solution: Fully dissolve 1g in 60mL pyridine, and dilute with 40mL acetonitrile, Fully dissolve 2g in 120mL pyridine, and dilute with 80mL acetonitrile See technical bulletin for details. |
| Sulfurizing: A 60 second sulfurization step time for DNA or a 360 second sulfurization step time for RNA is recommended. Sulfurization step must come before capping step to minimize phosphodiester formation. |
