High-Efficiency PurificationThe high affinity of fluorous-tagged oligonucleotides to fluorous solid supports has been exploited in a range of products useful for highly efficient purification of oligonucleotides. This high efficiency is especially useful in the purification of long oligonucleotides (50 - 100 mer) where recoveries of the order of 70-100% are routinely achieved. A fluorous version of CPR II has also been introduced for the simple purification of long oligonucleotide 5'-phosphates, with a potential use in gene construction.
dC-BRANCHERIn DNA diagnostics, the most common approach to the detection of a small amount of target is amplification of the target sequence. However, an alternative approach does exist -direct analysis of the target DNA by signal amplification. This technique requires the synthetic oligonucleotide to contain a primary sequence of interest complementary to the target attached to many identical copies of a secondary sequence to be used for detection. It is the detection of the many copies of the secondary sequence that serves to amplify the signal. The resulting branched oligonucleotide has been aptly described as comb-like, with the primary sequence in the handle and the secondary sequences being the teeth of the comb. The branches in the synthetic oligonucleotide are created using dC-Brancher Phosphoramidite. |
1-MethyladenosineModified nucleosides are found in large quantities in nucleic acids, especially RNA, and contribute to a wide variety of biological functions, some of which are essential and some detrimental. 1-Methyladenosine is especially interesting since it is an essential component when present in tRNA and is a mutation when found in DNA as the 2'-deoxynucleoside. Both the ribonucleoside and the 2'-deoxynucleoside are now available suitably protected for simple incorporation into RNA and DNA oligonucleotides. Sulfurizing Reagent IIA second sulfurizing reagent has been added to our product line of DNA synthesis reagents to complement our existing Sulfurizing Reagent (Beaucage Reagent). Sulfurizing Reagent II is stable in solution and is offered both in powder form and as a solution for use in popular commercial synthesizers. In addition to good performance in the synthesis of DNA phosphorothioates, Sulfurizing Reagent II demonstrates superior performance in the production of RNA phosphorothioates. ![]() Oligonucleotide CapsTwo new products for the synthesis of oligonucleotides allow the preparation of hybridization probes with increased affinity for complementary sequences. Both are phosphoramidites that can be readily introduced at the 5' terminus via automated DNA synthesis. These oligonucleotide caps favor the formation of stable Watson-Crick duplexes by stacking on the terminal base pair. Melting point increases of over 10 °C per modification can be realized for short duplexes. AP-dC (G-clamp) is a very important addition to the modified nucleosides that enhance hybridization since the AP-C....G base pair contains 4 hydrogen bonds, which makes the interaction much stronger than the regular C....G base pair with its 3 hydrogen bonds. The AP-dC modified base is also stable to conditions of oligonucleotide synthesis and deprotection. We envisage applications for G-clamp in the development of short, easily prepared oligonucleotides for use in highly specific single nucleotide polymorphism (SNP) and other in vitro diagnostic assays.
